Plant research reveals new role for gene silencing protein

ScienceDaily (Mar. 29, 2012) ? A DICER protein, known to produce tiny RNAs in cells, also helps complete an important step in gene expression, according to research on Arabidopsis thaliana. The expression of a gene, when an organism's DNA is transcribed into a useable product, requires activation via a promoter or an external trigger. Plant research to be published in Science helps to show that later stages of transcription are just as important. This is likely to apply to other organisms, including humans.

Termination is the final stage of transcription. Successful termination is dependent on DNA being transcribed into RNA with the correct sections, including a certain length tail.

Scientists at the John Innes Centre on Norwich Research Park have found that where effective termination through the normal mechanisms has not occurred, DICER-LIKE 4 (DCL4) steps in to tidy up. Without termination, transcription continues down the chromosome unchecked.

In this way, DCL4 plays a crucial and previously unknown role in transcription termination. It helps formation of the gene product. DCL4 is more commonly known to play a part in the opposite effect, gene silencing.

"DCL4 is a back-up to termination processes, helping a gene to be successfully expressed," said lead author Professor Caroline Dean from JIC, which is strategically funded by BBSRC.

The findings may help explain why gene silencing happens so often with transgenes. It was not known that so much attention should be given to the tail end of a gene.

"Our research shows that for successful expression the end of a gene is just as important as its beginning," said Dean.

When termination fails a lot of aberrant RNA is made -- this is degraded as part of a cell's quality control mechanism. This can have consequences for other sequences in the genome that match the aberrant RNA.

"If a gene ends badly, aberrant RNA will trigger silencing pathways," said Dean.

DCL4's ability to step in to rescue poor termination makes it important for both successful gene expression, a previously unknown role for it, and gene silencing.

The research was funded by JIC's strategic funding from BBSRC and by the EU research project SIROCCO, focused on silencing RNAs

A question of intelligence

Why are humans the smartest animals on Earth?

OUR intelligence, more than any particular behaviour or anatomical feature, is what distinguishes humans from the myriad other species with which we share our planet. It is a key factor in everything from our anatomy to our technology. To ask why we are intelligent is to ask why we are human; it admits no discrete answer. But let's ask it here anyway. Why are we, alone in nature, so smart?

Perhaps we are not. Maybe our anthropocentric conceit prevents us from fully appreciating the intelligence of other animals, be they ants, cephalopods or cetaceans. As Douglas Adams put it: "Man had always assumed that he was more intelligent than dolphins because he had achieved so much - the wheel, New York, wars and so on - whilst all the dolphins had ever done was muck about in the water having a good time. But conversely, the dolphins had always believed that they were far more intelligent than man - for precisely the same reasons."

So let's rephrase the question. There is a cluster of abilities that seems unique to humans: language, tool use, culture and empathy. Other animals may have rudimentary forms of these abilities, but they do not approach humans' sophistication and flexibility. Why not?

Some come closer than others. German psychologists say they have identified a chimp whose mental abilities far surpass those of its peers (see "Chimp prodigy shows signs of human-like intelligence"). Intriguingly, they go on to suggest that this might be because Natasha, the simian prodigy, exhibits strong social-reasoning skills, such as learning from others. These are the same skills to which the explosive development of human intelligence is increasingly attributed.

At least some chimps, then, may have the potential to evolve human-scale intelligence. But they haven't - at least not so far. Let's rephrase the question again. Why haven't all chimps evolved to be as smart as Natasha?

Some did, but a long time ago: our own ancestors. Somewhere in our evolutionary history, there were presumably similarly prodigious protohumans, produced by some accident of genetics or environment, whose greater intelligence gave them the edge over their less gifted peers. Today's chimp prodigies do not seem to profit from their intelligence in the same way. Their society and environment do not reward it as ours did.

So our ancestors may have been fortuitously placed to embark on the runaway cycle of biological and cultural development that led to modern, multitasking humans (see "Puzzles of evolution: Why aren't we more like chimps?") - and to a level of adaptability that allows us to adjust readily to changes in our environment, and even modify it to suit ourselves.

Yet we should beware of hubris. In the not-too-distant past, we shared the planet with distinctly different branches of the human family: the Neanderthals and Denisovans, perhaps the Flores and recently discovered Red Deer Cave peoples, and possibly many more. These hominins probably shared many of our mental abilities - and yet still found themselves unable to overcome their environmental challenges and so ultimately died out.

So a final question: does our intelligence really make us the rulers of the world? It's too early to say.

People With Autism May Be Better at Processing ... - Health News

MONDAY, March 26 (HealthDay News) ? People with autism have an enhanced ability to process information, which may explain the apparently higher-than-average percentage of people with autism who work in the information technology industry, British researchers say.

Along with this heightened capacity for processing information, people with autism are better able to detect information that is considered critical, according to the study, which appeared March 22 in the Journal of Abnormal Psychology.

Autism spectrum disorders include a range of neurodevelopmental disorders, all marked by difficulties with social and communication skills and repetitive behaviors.

People with autism have an increased ability to focus on certain tasks, but are also easily distracted by things such as flashing lights or particular sounds, it has been thought.

A University College London (UCL) researcher wondered if these conflicting characteristics might be due to a higher-than-normal ability to process information.

?Our work on perceptual capacity in the typical adult brain suggests a clear explanation for the unique cognitive profile that people with autism show,? Nilli Lavie, a professor at the Institute of Cognitive Neuroscience at UCL, said in a Wellcome Trust news release.

?People who have higher perceptual capacity are able to process more information from a scene, but this may also include some irrelevant information that they may find harder to ignore,? she said. ?Our research suggests autism does not involve a distractibility deficit but rather an information-processing advantage.?

Lavie and her colleagues tested the theory by giving 16 adults with autism and 16 adults without autism a task designed to assess their ?perceptual load capacity.? Both groups were successful at the task in the easier initial stages, but the adults with autism significantly outperformed those without autism as the task became more difficult.

?Our study confirms our hypothesis that people with autism have higher perceptual capacity compared to the typical population,? Lavie said. ?This can only be seen once the task becomes more demanding, with more information to process. In the more challenging task conditions, people with autism are able to perceive significantly more information than the typical adult.?

Lavie said the findings may help explain why people with autism spectrum disorders, such as Asperger?s syndrome, may excel in information technology and other careers that require intense concentration and the ability to process large amounts of information from a computer screen.

DAKAR, Senegal (AP) ? President Abdoulaye Wade conceded defeat to his former protege Macky Sall late Sunday, congratulating him several hours after polls closed when preliminary results showed the opposition candidate had trounced the 85-year-old incumbent.

Wade called Sall around 9:30 p.m. (2130 GMT) Sunday to congratulate him on his victory, state television reported. The move alleviated fears that Wade would attempt to stay in office after 12 years or would challenge the runoff results.

Even before Wade conceded, Sall's supporters began celebrating in the streets of the capital, singing and marching through downtown Dakar. Some even danced on the roofs of moving vehicles, and one man did a cartwheel amid the traffic near the Place de l'Independance.

Sociologist Hadiya Tandian said that Wade's concession washes away the wounds of a violent election season, which left at least six people dead and tarnished the country's reputation.

"This is a great victory for Senegal ? it shows the maturity of our democracy," Tandian said. "It shows that the Senegalese believe in their voter IDs, that a voter card can change something, can make a difference. It shows that our long democratic heritage continues to live in us day by day."

Senegal's democratic roots run deep, and many feared Wade would not concede defeat and in the process muddy Senegal's image as one of the only established democracies in western Africa.

Whereas most African countries began holding elections post-independence in the 1960s, the Senegalese first cast their ballots 164 years ago starting in 1848 when France gave its territory the right to elect a deputy to the French parliament.

At a midnight press conference at a Dakar hotel, Sall offered few details on the conversation he had with Wade earlier in the evening. Instead, he praised the voters and said he would be the president for all Senegalese.

"Tonight, a new era begins for Senegal," Sall told the hundreds of journalists and euphoric supporters who crammed into the venue to hear him speak.

There was no immediate comment from Wade himself, though his spokesman Serigne Mbacke Ndiaye confirmed the president's concession of defeat.

"Senegal, through a transparent election, just proved once again that it remains a great democracy, a great country," he said in a press release.

Wade first took office in 2000 after his predecessor conceded in a historic moment for Senegal. He easily won re-election in 2007, but has seen his popularity suffer amid soaring costs of living and unemployment. When he cast his ballot last month in the first round of balloting, some voters even booed him at the poll shouting: "Old man, get lost."

His image also was tarnished after he began giving an increasing share of power to his son Karim, who was derisively called "the Minister of the Sky and the Earth" after he was handed control of multiple ministries including infrastructure and energy.

Wade's reputation took a nosedive when he announced last year that he planned to run for a third term. For weeks leading up to last month's election, protesters calling for Wade to step down hurled rocks at police in demonstrations that paralyzed the capital's economic heart.

In recent weeks, images of Wade on campaign posters had their eyes scratched out. And his convoy was hit by rocks in the final days of the runoff campaign.

Marieme Ousmane Wele, 55, said she had voted for Sall because the rising prices of basic goods have made her life increasingly difficult.

"I sell cereal made from corn but the price of corn has really gone up. Now, I don't have many customers and it's becoming difficult to feed my own family," she said, as men sat nearby on plastic lawn chairs in the sand listening to news about the election on portable radios.

Others, though, praised Wade for the economic progress made during his 12 years in power. At a polling station in the suburb of Grand Yoff, Raymonde Semou, 64, said Sunday she personally credited Wade with helping two of her six children find work.

"Before, I had to sell grilled peanuts to feed my family and it was very difficult for me," she said.

Now, her employed sons have bought land to build a house, and she adds there is now electricity in her hometown in Senegal's restive southern Casamance region.

Sall, 50, a former prime minister who ran Wade's last campaign in 2007, is a geologist by training who worked for years under Wade. The two, though, had a subsequent falling out and during the campaign Wade referred to Sall as an apprentice who had not yet taken in "the lessons of his mentor."

Most simply spoke of change rather than Sall's credentials when explaining how they voted Sunday.

Dr. Johny Assane said he voted for Wade in 2000 but has since become disillusioned. While he says he is financially secure, he has seen how others have failed to benefit from Wade's leadership.

"The situation of my patients who come to get medicine in my office has really deteriorated," he said. "Everywhere there are children whose parents are finding it difficult to pay for their treatment and that shows me that the country is not working."
